Hypothalamic control of growth hormone release

The growth hormone–releasing hormone (GHRH) receptor is a class B G protein–coupled receptor enriched in somatotroph cells of the anterior pituitary. Educational interest in this receptor centers on how hypothalamic peptides initiate growth hormone release before downstream IGF-1 signaling unfolds.

What it is

The GHRH receptor binds growth hormone–releasing hormone and related analogs. Ligand binding activates Gs-coupled signaling that elevates cyclic AMP in somatotrophs, a key step toward growth hormone secretion.

Because GHRH tone helps set pituitary GH output, this receptor is a recurring reference point in research on growth hormone axis peptides.

How it works

GHRH engages the receptor on pituitary somatotrophs. Gs activation stimulates adenylyl cyclase, raising cAMP and protein kinase A activity. Downstream ion-channel and secretory-machinery changes support GH vesicle release.

Somatostatin and other modulators can oppose this drive, so GH output reflects a balance of stimulatory and inhibitory inputs rather than a single switch.

Biological role

In physiology, GHRH receptor signaling helps coordinate linear growth, body composition cues, and metabolic adaptations tied to the GH/IGF-1 axis. In research settings, selective agonists are used to probe receptor pharmacology and secretory dynamics.
